To characterize the properties of circulating mononuclear phagocytes in young normal humans, we studied blood mononuclear phagocytes from human subjects aged 1 to 35 years. The method employed allowed the separation of three mononuclear phagocyte populations, which were characterized by ultrastructural, biochemical, and functional criteria. Phagocytic activity and cytofluorimetric analyses demonstrated that the three circulating mononuclear phagocyte populations differed both in cell size and in their surface membrane and cytoplasmic constituents. One population was small and showed a low phagocytic activity. Phagocytosis was not inhibited by metabolic inhibitors, thus suggesting the existence of a nonmediated pathway for phagocytosis. Biochemical analysis of the phagocytes showed that they had a high phosphatase activity, which appeared to be mainly located in their plasma membranes. The second population was larger and contained dendritic cells which had a phagocytic activity, mainly dependent on temperature and involving a flocculent mechanism. These cells also had a high alkaline phosphatase activity. Biochemical analysis of the third population revealed that it was composed of monocytes which showed a high nonspecific esterase activity.
